EDTA tube, officially known as ethylenediaminetetraacetic acid tube, is usually marked by purple cap in routine blood examination. It's the most standard and basic type of blood collection tube in hospital laboratories.
Its core feature is the pre-added EDTA anticoagulant (usually EDTA-K2 or EDTA-K3) inside the tube wall. Its design aims to provide clinicians with high-quality anticoagulant whole blood samples for various critical hematology tests.

Working Principle

 

The anticoagulant effect of EDTA tubes is based on a chemical chelation reaction. Its core mechanism can be summarized into three key steps:

1.Chelated calcium ions: EDTA molecules have a unique structure, where the amino and carboxyl groups in the molecule act like "pincers" and form highly stable, soluble chelates with free calcium ions (Ca²⁺) in blood at a molar ratio of 1:1.

2.Blocking the coagulation cascade: Calcium ions (Ca²⁺) are essential cofactors at several key stages of blood coagulation, including the conversion of prothrombin (Factor II) to thrombin (Factor IIa) and the conversion of fibrinogen to fibrin. Once free calcium ions in the blood are effectively bound and depleted by EDTA, the entire endogenous and exogenous coagulation cascade cannot be initiated.

3.Achieving anticoagulation: Because the coagulation waterfall reaction is blocked, blood can be stably stored in a whole blood state inside the tube, effectively preventing the formation of fibrin clots. This protects the natural morphology and quantity distribution of blood cells (red blood cells, white blood cells, platelets), and provides reliable samples for subsequent analysis.

 

Additives' Differences in  Physicochemical Properties:

1.Chemical form: EDTA-K2 is a dipotassium salt, while EDTA-K3 is a tripotassium salt. This difference directly affects the pH and osmotic pressure of the aqueous solution.

2.Osmotic pressure effect: The osmotic pressure of K3 salt solutions is higher than that of K2 salts. This causes red blood cells in the K3 environment to more easily shrink due to water leakage (reduced volume), while K2 has a relatively smaller impact on red blood cell morphology and volume.

3.Dosage form and dilution effect: EDTA-K2 can be spray-dried and coated on the inner wall of the blood collection tube, avoiding the blood dilution effect of liquid anticoagulants and reducing the impact of osmotic pressure changes. K3 is mostly added in liquid form, resulting in a slight dilution effect.
