The COVID-19 antigen test kit is an in vitro diagnostic product that quickly determines infection by detecting characteristic proteins (antigens) on the surface of the virus.

The kit usually consists of three parts:

 

Test reagent card: The plastic cartridge is embedded with test paper strips and is the core testing component.

 

Sample extraction solution (buffer): Used to process collected samples, providing appropriate pH and ionic intensity to ensure normal antigen-antibody reactions.

 

Sampling swab: Used to collect nasapharyngeal swab or oropharyngeal swab samples.

Technical Principle

 

The reagent uses a double-antibody sandwich immunochromatography method. Simply put, the reagent contains two types of antibodies that can specifically recognize the coronavirus N protein (nucleocapsid protein): one labeled on colored particles, and the other fixed on the T-line (detection line) on the test paper. When the sample contains viral antigens, they sequentially bind to these two antibodies, forming an "antibody-antigen-antibody" complex, which clusters and develops color on the T-line, forming "two lines."

Why choose N protein as the detection target

 

The novel coronavirus contains four structural proteins: S, E, M, and N. Among them, N protein has the highest content in the virus and has fewer mutations compared to S protein. Choosing N protein as a target not only helps improve detection sensitivity but also reduces the risk of missed detection caused by viral variants.

Rapid Detection: 15-minute window period

Traditional nucleic acid testing requires amplification steps, taking at least 4-6 hours. Antigen detection uses immunochromatography principles: after the sample is dropped, the liquid flows from one end of the test paper to the other under the capillary pull of the nitrocellulose membrane, completing the entire reaction within 10-15 minutes. This "point-of-care testing" (POCT) concept expands testing scenarios from laboratories to communities, homes, and anywhere in need.

Easy operation: self-test-friendly design

The antigen test kits are designed to allow non-professionals to operate accurately. A typical operating process requires only three steps:

Sampling: Swab swab rotated inside the nasal cavity (at least 5 times per nasal cavity, no less than 15 seconds)

Processing: Insert the swab into the extraction solution and rotate and squeeze to ensure the sample and buffer mixture are fully mixed

Testing: Add 4 drops of treatment solution to the sample well of the reagent card, read the result after 15 minutes
